▶Abctract
CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) system, a revolutionary genome editing tool known as third-generation molecular scissors has been developed and, since its first application to eukaryotic cells including human cells, dramatic academic achievements in the field of genome editing have accumulated over the short period of time. CRISPR system consists of two components: a specific guide RNA (gRNA) and a non-specific CRISPR-associated endonuclease protein from Streptococcus pyogenes (Cas9). Originally discovered as a form of prokaryotic immunity, once directed to a specific DNA sequence target by complementary gRNA, Cas9 causes a double-strand break (DSB) in its target loci. After the cut, CRISPR genome editing system relies on the cell’s natural repair mechanisms either generating Indels or incorporating desired edits at the genome. This core working mechanism enables us to carry out various molecular genetic applications in the genome. Among many biological applications, it has a great potential to shorten the generation of crops/livestock varieties in terms of pin-point gene correction. Various studies related to this novel molecular breeding in various parts of the world are emerging as a hot topic, and we are poised to make food items made from "genetically corrected" ingredients based on such technologies in the near future. Beside genetic knock-out and knock-in, many other CRISPR derivatives are now being discovered and developed. Today's seminar will help you to understand how this CRISPR system works: by understanding how it's done, what is different and what potentials it has in agricultural molecular breeding. In addition, Adenine Base Editor (ABE), a recent alternative to Cas9 and the many other modified applications will be discussed for their potential contribution to the field of biology, biotechnology and biomedicine through a couple of examples
