 Aging process confers multiple alterations across from molecular and physiological level. Among molecular alteration is loss of porteostasis upon aging. However, underlying mechanisms leading to age-induced loss of proteostasis in muscle, and proper treatment is largely lacking. In this study, we provide evidences that ceramide and ceramide producing pathways are enhanced in muscles of aged mouse, C.elegans and human. Along with this, we also showed increased protein aggregates in aged muscles. Based on these observations, we examined therapeutic effect of limiting ceramide levels on aggregate reduction and muscle function improvements. Treatment of myriocin, an inhibitor of SPT, which is the most upstream enzyme for ceramide de novo synthesis, into the aged mice reduced age-induced aggregate accumulation in skeletal muscles. Additionally, myriocin also improved muscle functions and glucose metabolism. In line with this, myriocin improved mobility, and reduced aggregates in aged C. elegans. To make it clear that myriocin improves muscle function via reduction of protein aggregates, we treated myriocin into human A1-42 expressing C. elegans (GMC101). Myriocin enhanced mobility and lifespan of GMC101, and reduced aggregate levels. In a mechanistic perspective, we showed that beneficial effects of myriocin are dependent on the induction of mitophagy.
